Comparison of HPLC-Chip/MS with conventional nanoflow LC/MS for proteomic analyses

چکیده
Nanoflow LC/MS is commonly used for protein identification and biomarker discovery, but setting up and maintaining the intricate LC plumbing can be a challenge. The Agilent HPLC-Chip represents breakthrough technology that integrates nanoflow HPLC columns, associated connections, and a nanoelectrospray emitter on a single, small, reusable microfluidic chip. The HPLC-Chip and a companion HPLCChip/MS interface afford much better ease-of-use than traditional systems. This Application Note demonstrates performance of the HPLC-Chip/MS versus a state-of-the-art conventional nanoflow LC/MS. A direct comparison shows that the HPLC-Chip/MS system produces better chromatography and sensitivity, leading to more identified peptides and proteins. Application Note Martin Vollmer Christine Miller Georges L. Gauthier cerevisiae). This work will illustrate that the chip format delivers reduced sample losses and lower delay volumes, thus producing uncompromised chromatographic performance and more identified peptides and proteins. In sum, the HPLC-Chip/MS system provides better results than a state-of-theart conventional system, with much better ease-of-use and reliability.

BACKGROUND The identification of interacting proteins within protein complexes is key to understanding the transduction and regulation of cell signaling pathways, and is also a useful tool for identifying novel disease markers. Immunoprecipitation of protein complexes followed by liquid chromatography-tandem mass spectrometry (LC-MS/MS) has been used to identify targets that bind to a protein o...
متن کاملChip-based nLC-TOF-MS is a highly stable technology for large-scale high-throughput analyses.
Many studies focused on the discovery of novel biomarkers for the diagnosis and treatment of disease states are facilitated by mass spectrometry-based technology. HPLC coupled to mass spectrometry is widely used; miniaturization of this technique using nano-liquid chromatography (LC)-mass spectrometry (MS) usually results in better sensitivity, but is associated with limited repeatability. BACKGROUND Detection limit challenges associated with measuring low-abundance protein biomarkers can be addressed with hybrid immunoaffinity-mass spectrometric assays, such as antipeptide antibody capture followed by liquid chromatography/tandem mass spectrometry (LC-MS/MS). Popular assay formats use magnetic bead-based immunoaffinity enrichment and nanoflow LC-MS/MS or high-flow immunoaffinity...
